Deficiency of macrophage migration inhibitory factor gene delays healing of the medial collateral ligament: a biomechanical and biological study.

چکیده
The role played by macrophage migration inhibitory factor (MIF) in the process of wound healing is controversial. Besides, there have been no reports that investigated the expression or the role of MIF in the repair process after ligament injury. In this study, we hypothesized that the deficiency in MIF gene might delay ligament healing in mice. The aim of this study was to clarify this hypothesis using MIF gene-deficient mice (MIFKO) and murine model of injury to the medial collateral ligament (MCL). Biomechanical testing showed that the levels of mechanical properties were significantly lower in MIFKO than in wild-type mice (WT) on day 28 after injury. Levels of matrix metalloproteinase (MMP)-2 and -13 mRNA in the healing tissue were significantly lower in MIFKO than in WT on day 28 and on day 7, respectively. Histologically, healing tissues in MIFKO exhibited prolonged hypertrophy, poor vascularity, and prolonged increase in cell number compared with those in WT. Taken together, it was suggested that MIFKO exhibited delayed healing of the MCL, which might be caused by lower mRNA expression of MMP-2 and -13.

INTRODUCTION: Ligament healing is composed of several phases, including acute inflammation and reaction phase, repair and regeneration phase, and remodeling phase. At the molecular level, these phases are regulated by a complex interplay among inflammatory cells, fibroblasts, and the molecules, such as inflammatory cytokines, proteinases, extracellular matrices, and etc, which are produced by t...

Effect of Low- Power Helium-Neon Laser Irradiation of Healing of Incisized Medial Collateral Ligament of Knee in Rars
Purpose: The aim of the present study is to investigate the effects of LPL on healing of incisized medial collateral ligament (MCL) of knee of rats. Materialsand Methods:MCL of right hind limb of 30 rats were transversly incisied. LPL (Helium- Neon, Continuose, output 10mW) were exposed (10 mJ/cm2 and 1.2 J/cm2) daily to group 1& II (experimental groups) respectively. In control group MCL were...
متن کاملCombined use of bFGF and GDF-5 enhances the healing of medial collateral ligament injury.
Basic fibroblast growth factor (bFGF) and growth and differentiation factor (GDF)-5 stimulate the healing of medial collateral ligament (MCL) injury. However, the effect of isolated and combined use of bFGF/GDF-5 remains still unclear. We investigated cellular proliferation and migration responding to bFGF/GDF-5 using rabbit MCL fibroblasts. Rabbit MCL injury was treated by bFGF and/or GDF-5 wi...

INTRODUCTION: Anterior cruciate ligament (ACL) and medial collateral ligament (MCL) are the major stabilizers of knee joint. The deficiency of these ligaments could result in the early development of osteoarthritis. The cellular activities responding to growth factors have important roles in ligament healings. ACL has a poor healing potential rather than MCL after injuries. To accelerate the li...
